Environmental genotoxins and metabolic by-products generate DNA lesions which can cause genomic instability and drive carcinogenesis. To ensure genomic integrity, cells employ mechanisms that convert signals generated by stochastic DNA damage into organized responses including activation of repair systems, cell cycle checkpoints and apoptotic mechanisms. We have identified a novel, DNA damage response (DDR) pathway that initiates in response to transcription-blocking DNA damage and activates the master DDR coordinator ATM, through mechanisms involving spliceosome remodeling and formation of potentially genotoxic R-loops. The cellular DNA damage response has received considerable attention as therapeutic target for cancer and a full understanding of the mechanisms underlying this novel DDR-related pathway involving R-loops might represent exceptional opportunities to achieve better understanding of cancer biology and to develop more precise cancer therapies.

The PhD candidate will participate in this novel and exciting line of research, focusing on the elucidation of the exact molecular mechanism of DNA damage signaling by R-loops and how the latter are resolved to prevent further genomic instability. To this end, the PhD candidate will employ a multidisciplinary approach, using various biochemical and DNA repair assays, live cell imaging and quantitative proteomics.

We are looking for a highly motivated and talented candidate that holds an MSc degree in Biomedical Science, Molecular/ Cellular Biology, Biochemistry or related field with a basic knowledge of DNA repair pathways. Experience with biochemical assays, standard molecular biology techniques, live cell imaging and proteomic tools will be a strong advantage. Excellent writing and presentation skills in English are recommended. As you will work in dynamic international research we need a team-player who is able to combine independent thinking with a collaborative attitude.

Being able to present a certificate of good conduct, a valid proof of identity, diploma's and/ or relevant registration such as BIG/ RGS are conditions for the appointment.

You will receive a temporary position for 4 years. The gross monthly salary is € 2.279 ,- in the 1st year and increases to € 2.919,- in the 4th year (scale OIO). The terms of employment are according to the Collective Bargaining Agreement for Dutch University Medical Centers (CAO UMC).
