Loss of vision changes the lives of many children and young adults dramatically. One of the most frequent causes of blindness early in life is due to the loss of USH2A function. Up until recently, it was not possible to use generic gene therapy vectors to fight retinal degeneration for large genes such as USH2A. Now however, we employ high-capacity adenoviral (HC-AdV) gene therapy vectors that can incorporate the USH2A gene and target photoreceptors. In this project, the vector will be used to develop generic USH2A gene augmentation therapy as well as generic USH2A CRISPR-CAS gene editing therapy. As PhD candidate, together with your colleagues, you will perform proof of concept studies by testing both of these vectors in cultured human induced pluripotent stem cells (hiPSC-), derived retinal organoids and hiPSC-retinal pigment epithelium. During the project, you will develop skills in hiPSC retinal organoids and retinal biology, viral gene augmentation, retinal organoids-on-a-chip technology, and CRISPR-Cas gene editing therapy vector development for future clinical studies. You communicate with team members, scientists, and peers at (inter-)national scientific meetings, and perform mechanistic and translational studies.
We are searching for a highly motivated, creative, and proactive PhD candidate who works well independently. You are also an eager communicator, and can transfer knowledge you have acquired well. We accept international students, but a MSc degree in Life Sciences is required, together with sufficient laboratory experience. Furthermore, you have hands-on experience with cell biology and molecular biology techniques such as cell culture (preferably stem cells such as human iPSC or EC), gene expression systems (preferably viral vectors), molecular cloning, qPCR, western blotting, (immuno-)histochemistry, and (confocal) microscopy. Lastly, you have excellent communication skills in English.
You will be offered an employment contract for the production of a PhD thesis for the duration of the program with a maximum of four years. The salary amounts to € 2,495 gross per month in the first year, up to a maximum of € 3,196 in the last year with fulltime employment (scale Pro, CAO UMC)
